抄録: The effect of five different antibiotics on chemotaxis of human polymorphonuclear (PMN) leukocytes was studied in an in vitro system using an agarose plate method. PMN leukocytes were collected from 30 healthy human volunteers (between 20 to 79 years of age), and they were incubated with carbenicillin, cephaloridine, gentamicin, tetracycline and chloramphenicol. Quantitation of chemotaxis is done by measuring in centimeters the linear distance the cells have migrated from the margin of the well toward the chemotactic factor. Random mobility is represented by the distance the cells have migrated from the well margin toward the control medium. The results were summarized in the followings: (1) In higher age more than 50 years old, the values of chemotaxis were 70 to 80% compared with those in the age group under 49 years old, though there was no significant influence of age on drug-induced suppression of chemotaxis by statistical analysis. (2) There was almost no effect of the betalactam antibiotics (CBPC, CER) on the chemotaxis. (3) Tetracycline of the therapeutic blood level (3.12 ug/ml) appeared to inhibit PMN leukocyte chemotaxis, whereas there was a tendency to fail to suppress leukocyte chemotaxis in very high concentration (500 µg/ml) of tetracycline. (4) Chloramphenicol in concentrations of 3.12 µg/ml, 25 µg/ml, 100 g/ml and 500 µg/ml inhibited the PMN leukocyte chemotaxis. (5) Gentamicin seemingly suppressed PMN leukocyte chemotaxis at the high concentrations. An increasing inhibition was seen with the higher concentrations. The followings were postulated: (1) The increased production of lactic acid by leukocytes in the presence of 500 µg/ml of tetracycline may be a manifestation of a metabolic effect of the drug, and the low levels of tetracycline may effect to microfilaments of leukocyte. (2) In the presence of chloramphenicol, the increased production of intracellular cyclic AMP in leukocytes restrict an intact system of cytoplasmic microtubules. (3) Gentamicin has a character such as chemoattractant substance.
