Abstract: | A highly emissive thio-analogue of deoxyguanosine triphosphate, [th]dGTP, was synthesized and enzymatically incorporated into DNA through primer extension and PCR amplification. The straightforward amplification of [th]dG-labeled DNA by natural polymerases and simple addition of [th]dGTP to the standard PCR mix was demonstrated. The present results will facilitate the broad application of long fluorescent DNA. |
