Abstract

First, serum acid phosphatase from healthy adult men was studied by column chromatography with stepwise elution technique using DEAE cellulose. Three distinct peaks of protein (Fr. I, II and III) were obtained and acid phosphatase activity was exhibited in each fraction. When seminal plasma was added to the same normal serum chromatography demonstrated extremely increased enzymatic activity on Fr. II and III, which were suggested to be of prostatic origin. Chromatography of the extracts obtained from kidney, liver, spleen and red blood cells suggested that the enzyme contained in these organs was distributed to Fr. I and that in red cells to Fr. II. The sera of patients with prostatic cancer, benign prostatic hypertrophy and chronic renal insufficiency were then chromatographed. Serum from patients with benign prostatic hypertrophy and chronic renal insufficiency showed the pattern of distribution of acid phosphatase in each fraction similar to that of normal serum. However, the enzymatic activity of Fr. III of patients with prostatic cancer revealed 12.6% of total activity, while those of the other cases represented below 2 %.
