Rifampicinに関する基礎的ならびに臨床的検討

Abstract

This report deals with laboratory and clinical assessment of Rifampicin on urinary tract infection. The results obtained were summarized as follows. 1) MIC of Rifampicin (RFP) against 183 strains of bacteria which were isolated from urinary tract infection during the recent four month period in our clinic, was determined by plate dilution method. It was discovered by this determination that RFP had excellent antibacterial activity against Staphylococcus aureus and Staphylococcus epidermidis and that MICs were less than 0.1mcg/ml in great majorities of these two species. E. coli was proved to be less sensitive to RFP, and MICs were distributed from 3.13 mcg/ml to 200 mcg/ml. Other Gram-negative rods such as Proteus and Pseudomonas seemed to have much the same sensitivity as E. coli. MICs of Enterococcus were situated between 1.56 mcg/ml and 50 mcg/ml 2) Rat tissue assay of RFP was performed 1, 2, 4, 6 and 8 hours following a peroral administration of 10 mg per kg body weight. This tissue assay revealed that the liver had the highest activity and reached a peak of H.5 mcg/ml six hours after the administration. Activity of other organs was in order as follows-serum, kidney, lung and spleen. 3) Serum concentration of RFP following a single administration of 450 mg in two healthy adults reached a peak level of 3.5 mcg/ml in average four hours later. Urinary recovery rate during the first six hour period came to 6.42% in average in three healthy males who had taken 450 mg of RFP. 4) Of twelve patients who suffered from acute simple cystitis, nine were effectively treated by peroral administration of 450 mg a day for three days. No significant side-effects were encountered in all of them.
