Abstract: | Six cases of healthy adult volunteer were employed for the study and divided into two groups. Three grams of cephalothin dissolved in 100 ml of sterile saline for injection was given to the one group by 1 hour drip infusion. The other was given 3 g of cephalothin dissolved in 500 ml of the saline by 2 hours drip infusion. Serum level and urinary excretion of the drug were measured by thin layer cup method using Bacillus subtilis A TCC 6633 as a test strain. A peak of serum level obtained by 1 hour drip infusion reached higher than that obtained by 2 hours infusion. Area under the plasma concentration time curve obtained by 1 hour drip infusion seemed to be wider than that by 2 hours infusion at 50 µg/ml. Urinary excretion rate up to 6 hours after the injection was 78.9% and 76.9% respectively. However, the rate up to 2 hours was higher in the group injected for 1 hour than 2 hours. Neither adverse side effect nor abnormal laboratory data was noticed after the administration of 3 g of cephalothin. |
