ABPC,CEZ,KMおよびOLMの膀胱壁移行に関する実験的研究

Abstract

Studies were performed on the movement of ABPC, CEZ, KM or OLM through the isolated urinary bladder of the rat. In advance of the studies, mechanography of the isolated bladders was recorded, and it was found that there were no changes on the mechanograms when the bladders were perfused with oxygenated Krebs' solution alone or the solution containing 60 mcg/ml of ABPC, CEZ, KM or OLM or !20 mcg/ml of KM. It was also found that oxygenated TC-Medium 199 (Difco) solution caused decrease followed by augment in tonus and contractibility of the smooth muscle of the bladder. In the studies on the movement of the antibiotics in the direction of from the inside of the bladder outwards, the bladders were filled with 0.7 ml of 60 mcg/ml of the antibiotic in PBS and perfused with oxygenated Krebs' solution. The bladders were divided to two groups. One group was used to measure the concentration of the antibiotic after 3 hours and the other after 6 hours. In the studies on the movement in the opposite direction, the bladders containing 0.7 ml of PBS in them were perfused with 40 mcg/ml of the antibiotic in oxygenated Krebs' solution. The concentrations of the antibiotics were measured 2 and 4 hours later. The series of the experiments were conducted at 37°C and at constant flow of 10ml/min of the perfusate. Any of four types of the antibiotic moved more rapidly into the inside through the bladder wall than did out of the bladder. Velocity was in following order; ABPC<CEZ≒OLM<KM in the direction of from the inside of the bladder outwards, KM<ABPC≒CEZ≒OLM in the direction of from the outside of the bladder inwards.
