dc.description.abstractQuantitative measurement of mitochondrial DNA (mtDNA) has been very troublesome, because its content in total DNA was very low and quantitative separation of the mtDNA from nuclear DNA (nucDNA) was almost impossible. By using the technique for cloning cells carrying the mtDNA sequences, specific detection of mtDNA became possible in the presence of large excess of nucDNA by DNA-DNA hybridization. For this purpose, we prepared mtDNA probes labeled with a non-radioactive substrate, which made the procedure easier and increased possibility of application. This method revealed that the contents of mtDNA in rat liver strikingly decreased with aging.en
