抄録: | Why the sera of cancer, inflammatory disease and hepatic disease show the lower wave than normal? As already explained repeatedly in the previous reports, one of the factors for this phenomenon is due to the decreased content of serum protein accompanied with the disease. In the last report the clinical correlation of wave height to the A/G ratio was discussed. The clinical investigation was continued by the following modified method. In order to avoid the quantitative factor, each serum, containing different amount of protein, was adjusted to the definite concentration in the final cobaltous mixture (0, 15%), using the procedure of CuSO4 gravimetric method. Further, the temperature in the experiments was always kept constant (16℃). Comparing the wave-height obtained with the value of A/G ratio, we found that at least in the liver disease the relationship between these two values was very intimate, especially so far as the denaturated sera were concerned (Table I). In order to ascertain this results, we analysed the protein fractions, which had been isolated by various methods, such as those of ethanol-fractionation in -5℃ or of the salting-out by Na2SO4, and further the albumin crystal of horse serum (Mc. Meekin). With these samples, the relation of the wave-height-to its protein concentration was studied both in natural and in denaturated states. As seen from Fig. I and 2, the albumin revealed higher wave than the globulin and- when denaturated, the wave from albumin became very high, wheras the wave from globulin showed almost the same. The wave-forms were also characteristic to the nature of proteins, i. e. to the facts that either the fraction is albumin or globulin, the state is natural or denaturated, and the origin is human or horse (Fig. 3). From these facts it became apparent that the wave from the serum protein showed a combined wave of albumin and globulin, so that polarographic test for serum protein (the first reaction of Brdička or the digest test of O.H. Müller) represents essentially the test in regard to the A/G ratio. |
